2 March 2022 FOCALS microscopy: a label-free single-source multimodal optical system for holistic characterization of dynamic living samples

Abstract
We introduce FOCALS (Fast Optical Coherence, Autofluorescence Lifetime imaging, and Second harmonic generation) microscopy capable of generating NAD(P)H fluorescence lifetime, second harmonic generation (SHG), and polarization-sensitive optical coherence microscopy (OCM) images simultaneously and equipped with closed-loop sensorless adaptive optics (AO). Multimodal imaging generates quantitative metabolic and morphological characterization of biological samples and the complementary contrasts improve the overall specificity of our setup. We highlight the versatility of our setup to image seconds-scale dynamics of in vitro mammalian cells, ex vivo tumor and neural environments, and in vivo skin imaging with sub-micron resolution simultaneously.
