Mitochondrial outer membrane permeabilization (MOMP) is a decisive step in the execution of cell death, and MOMP is often altered in cancer cells. MOMP could provide an early measurement of cancer treatment efficacy, and a method to understand relationships between mitochondrial function and cell death. Cationic dyes such as TMRE label mitochondria in a membrane potential dependent manner and provide insight into mitochondrial function in living cells. Currently, there are no easy to use open-source analysis methods to rapidly quantify bulk mitochondrial membrane potentials on a per cell level. Here, we present a custom CellProfiler pipeline to isolate TMRE intensity in both the mitochondria and nuclei of cells for rapid cell level quantification of bulk membrane potential.
